Custom Synthesis

We can perform custom organic syntheses of bio-active compounds, metabolites, active pharmaceutical ingredients and stable-isotope labelled compounds at a competitive price. All synthesized compounds are subjected to rigorous quality control, including demonstrations of purity and identity by HPLC chromatography, mass spectrometry, nuclear magnetic resonance and elemental analysis. Analytical Chemistry

We are able to evaluate the quality of medical preparations, active substances, raw materials and intermediate products, and can also conduct stability studies on these species. At the client's request, we can also test, design, and validate analytical methods.

Our equipment enables us to carry out standard and special physical and chemical analyses, including liquid chromatography, gas chromatography, liquid chromatography-mass spectrometry (LC-MS), infrared spectroscopy, titration and a number of other analytical assessments. Our experts have extensive experience of working under good manufacturing practices in the pharmaceutical industry.

Kinase Selectivity Screening

We can test the selectivity of a compound against panels of purified human protein kinases. Mis-regulation of kinase activity plays a major role in many diseases. However, because many kinases have similar ATP binding pockets, one of the biggest challenges when developing a kinase inhibitor is the optimization of its selectivity. Kinase inhibitors must therefore undergo thorough selectivity profiling. All our assays are conducted using ATP labeled with 33P with an ATP concentration equal to the target enzyme's Km. The panel contains over 100 protein kinases expressed in Escherichia coli or Spodoptera frugiperda cells. Each assay is performed in duplicate. The turnaround time is usually 2 weeks, and screeening is conducted once per month. We screen in two standardized formats:
  • at a single fixed concentration against the whole panel of kinases
  • by determining an IC50 value using a 10 point dose-response curve against any one kinase in the panel

Cellular Assays

We offer screening services to determine the effects of your compounds or extracts on the cellular level. Our repository contains up to 80 cell lines, including adherent and suspended cancer cells, endothelial cells, untransformed fibroblasts and immortalized hepatocytes. We have established a panel of simple assays that are particularly suitable for anticancer drug candidates, including tests for:
  • in vitro angiogenesis (cell migration and tube formation by HUVECs incl. quantitative image analysis)
  • apoptosis (subdiploid population, caspase fragmentation and activity, PARP1 cleavage)
  • cell cycle inhibition (PI and DAPI staining, BrdU incorporation, expression of cyclins or phospho-histone H3)
  • proliferation and cytotoxicity (MTT, Calcein AM, Alamar Blue, clonogenic/colony formation assay)
  • DNA damage (expression of ATM, CHK1, CHK2, SMC1, p53, gamma-H2AX)

Custom Antibodies

We are able to develop monoclonal and polyclonal antibodies in mice and rabbits, respectively. Our services can include antigen peptide design, synthesis and conjugation to BSA or KLH, immunization and boosts, serum testing, ELISA titration, final purification (protein A or G), and delivery of test samples, according the needs of each individual customer. We also have the expertise to produce antibodies against small organic molecules such as plant hormones or secondary metabolites. We can develop ELISA tests, characterize antibodies by immunoblotting, immunoprecipitation, immunohistochemistry, or immunofluorescence, and conduct affinity purification using antigen columns.

In addition, we can produce antibodies in a hybridoma cell line, which are secreted into the fluid ascites. This is done by injecting the hybridoma cell line into the peritoneal cavity of mice and harvesting the antibody-containing fluid.
