JNJ-7706621

JNJ-7706621 is a potent cell cycle inhibitor that targets several cyclin-dependent kinases (CDK) and Aurora kinases. It has IC50 values of 9 and 11 nM for CDK1/Cyclin B and aurora A, respectively, and blocked the growth of several different types of tumor cell in vitro ten times more effectively than it inhibited the growth of normal human cells. At low concentrations, JNJ-7706621 slowed cell growth; at high concentrations, it was cytotoxic. Flow cytometric analysis of cellular DNA content showed that JNJ-7706621 delayed progression through G1 and arrested the cell cycle in the G2-M phase. Additional cellular effects due to Aurora kinase inhibition included endoreduplication and inhibition of histone H3 phosphorylation.
