Tacrolimus (FK-506)

Tacrolimus is a potent immunosuppressant that is used after allogeneic organ transplantation to reduce the the risk of organ rejection. It disrupts signaling events mediated by calcineurin (Ca-dependent phosphatase) in T lymphocytes. Its mechanism of action involves the formation of a molecular complex with the intracellular FK506-binding protein-12 (FKBP12), thereby acquiring the ability to interact with calcineurin and to interfere with its access to and dephosphorylation of various substrates. The knows substrates of calcineurin involved in induced immunosuppression include the nuclear factors of activated T cells (NFAT).
